Pds5 proteins modulate the behavior of cohesins to ensure the proper division of cells -- Understanding the regulation of cohesins can improve diagnosis and treatment for some cancer patients or those suffering from Cornella de Lange Syndrome

Cohesins are protein complexes that join the two copies of each chromosomecalled sister chromatidsto ensure that they are shared fairly between the daughter cells during cell division. In this way, each daughter cell receives exactly the same genetic information from the parent cell.

Pds5 is a protein associated with cohesins; it binds cohesins along different chromosome regions. In vertebrates there are two variants of Pds5, Pds5A and Pds5B, not very well characterised to date. Scientists from the Spanish National Cancer Research Centre (CNIO), led by Ana Losada, from the Chromosome Dynamics Group, have discoveredby using genetically-modified mice (knock-out mice for Pds5A and Pds5B) that the two Pds5 variants are not equivalent, as both are necessary for cell proliferation and for embryo development to take place correctly.

The results, published today in the online version of The EMBO Journal, contribute to improving our understanding of how Pds5 proteins modulate the behaviour of cohesins, either by stabilising or destabilising the binding of cohesins to the chromosomes.

CLINICAL IMPLICATIONS OF THE STUDY

Illnesses such as Cornella de Lange Syndrome, which affects 1/30,000 newborns and which is characterised by serious physical abnormalities and cognitive impairments, have their origin in the abnormal function of cohesins. Cohesin mutations have recently been identified in some types of tumours, such as bladder cancer or acute myeloid leukaemia (AML). "If we understand the function and regulation of the cohesins, we can improve the diagnosis and treatment for affected patients", says Losada.

"In this context, we have seen that both forms of Pds5 stabilise the binding of cohesins to the arms and distal regions of chromosomes, whilst only Pds5B does so in the centromeres the chromosomal regions that are most critical for chromosome separation during cell division", explains Losada.

But Pds5 proteins not only stabilise cohesins, they can also have the opposite effect when they bind to the Wapl protein. "In this case, Pds5 contributes to destabilising the binding of cohesins to chromosomes, which can sometimes be important for activating essential genes at the right moment, as well as for allowing an efficient separation of the sister chromatids during cell division".

Conservative Group Backs Challenge To 'Liberal' McConnell

Days after Senate Minority Leader Mitch McConnell helped negotiate a deal to end the government shutdown and raise the debt ceiling, a prominent conservative group endorsed his primary challenger.

The Senate Conservatives Fund officially backed Tea Party favorite Matt Bevin Friday in Kentucky's 2014 Senate race, arguing he is "a full-spectrum conservative with the courage to stand up to the Washington establishment" while McConnell "has a liberal record and refuses to fight for conservative principles."

SCF has had McConnell in its cross hairs for months. In mid-September, the group launched a television advertising campaign attacking McConnell for not doing enough to defund the Affordable Care Act. It also ran radio ads in August calling on McConnell to oppose funding for Obamacare.

Bevin could use the financial support from SCF, which raised more than $16 million during the 2012 elections, if he hopes to pull off the upset. He brought in just $220,000 from July to September, while contributing $600,000 of his own money to the campaign. McConnell raised nearly $2.3 million during the same period and has more than $10 million on hand.

SCF, which has ties to former South Carolina senator and current Heritage Foundation President Jim DeMint, has made a name for itself supporting Tea Party challengers over Republican incumbents and establishment-backed candidates in Senate primary races.

The group appears to be revving up for the 2014 cycle with the shutdown now in the rearview mirror. On Thursday, the group backed Mississippi state Sen. Chris McDaniel over six-term GOP incumbent Sen. Thad Cochran.

One of SCF's endorsements in 2012 went to Ted Cruz, who has become the leader of the fight to defund Obamacare in Washington, in his primary campaign against Texas Lt. Gov. David Dewhurst.

September Jobs Report To Come Out Tuesday

Following the economy can be confusing.

But at least one thing has long been certain: the Bureau of Labor Statistics will release its monthly jobs report at exactly 8:30 a.m. on a Friday.

Next week, Tuesday will feel like a Friday.

That's because late Thursday afternoon, the BLS updated its post-shutdown schedule for data releases. The new schedule shows that the long-delayed and much-anticipated September employment report will come out on Tuesday.

The original release date was Friday, Oct. 4. But that jobs report got caught up in the government shutdown. Now it will be released Tuesday, Oct. 22.

And the October report, originally scheduled for release on Nov. 1, will be pushed to Nov. 8. That's late, but at least it's a Friday. TGIT just doesn't sound right.

England and defending champion Spain qualified for the World Cup on Tuesday night along with Russia and Bosnia-Herzegovina, which earned its first berth as an independent nation.

Three more spots were at stake in late games in South and Central America, finalizing 21 of the 32 slots for the field in Brazil next June.

Wayne Rooney scored in the 41st minute and Steven Gerrard in the 88th to give England (6-0-4) a 2-0 win over Poland at London's Wembley Stadium and first place in Group H by one point over Ukraine (6-1-3). The Three Lions qualified for their 14th World Cup and fifth in a row.

Spain (6-0-2) won Group I with a 2-0 victory over visiting Georgia on goals by Alvaro Negredo in the 26th minute and Juan Mata on Juan Mata in the 61st. France (5-1-2) was second and will be in the playoffs despite defeating visiting Finland 3-0 on goals by Franck Ribery and Karim Benzema around Joona Toivio's own goal.

Cristiano Ronaldo's Portugal, Greece, Ukraine, Romania and Iceland also finished second in their groups, winding up in Monday's draw for the eight-team European playoffs next month along with Sweden and Croatia. The four playoff winners also will earn berths.

Denmark (4-2-4) had the worst record of the nine second-place teams and missed out.

Bosnia (8-1-1) won 1-0 at Lithuania on Vedad Ibisevic's 68th-minute goal to win Group G on goal difference over Greece (8-1-1), which beat visiting Liechtenstein 2-0 as Dimitris Salpingidis scored in the seventh minute and Giorgos Karagounis added a goal in the 81st.

Russia (7-2-1) won Group F with a 1-1 tie at Azerbaijan as Roman Shirokov scored in the 15th. Portugal (6-1-3) finished a point back after defeating visiting Luxembourg 3-0 on goals by Silvestre Varela, Nani and Helder Postiga.

Later Tuesday, Ecuador, Chile and Uruguay competed for South America's final two automatic berths, with Uruguay and Venezuala having a chance to finish fifth — which earns a playoff against Jordan.

In North and Central America and the Caribbean, Honduras was three points ahead of Mexico for the region's last automatic berth, and Mexico was three points ahead of Panama for fourth playoff and a playoff against Oceania champion New Zealand. The U.S., which clinched last month, was at Panama, while Mexico was at Costa Rica, and Honduras at Jamaica.

Carol Burnett Receives Top US Humor Prize In DC

WASHINGTON (AP) — When Carol Burnett launched her namesake variety show in the 1960s, one TV executive told her the genre was "a man's game." She proved him wrong with an 11-year run that averaged 30 million viewers each week.

On Sunday, the trailblazing comedienne will receive the nation's top humor prize at the Kennedy Center for the Performing Arts. Top entertainers including Julie Andrews, Tony Bennett, Tina Fey and Amy Poehler will pay tribute to Burnett as she receives the Mark Twain Prize for American Humor.

The show will be taped and broadcast Nov. 24 on PBS stations.

In an interview, Burnett said she was drawn to comedy after realizing how it felt to make people laugh. She went to UCLA with plans to become a journalist, but she took an acting course that put her on stage in front of classmates.

"I played a hillbilly woman, and coming from Texas ... it was real easy for me," she said. "I just made my entrance, and I said, 'I'm Baaack.' Then they exploded."

"I thought whoa! This feels good," Burnett said. "I'd never been that popular in school. ... All of a sudden, it just opened up to me."

Few women were doing comedy when Burnett set her sights on New York. She caught a break when she was spotted by talent bookers from TV's "The Ed Sullivan Show" and was invited to perform her rendition of "I Made a Fool of Myself over John Foster Dulles."

Almost immediately, Burnett transformed Dulles, the former secretary of state, "from a Presbyterian bureaucrat into a smoking hot sex symbol," said Cappy McGarr, the co-creator of the Mark Twain Prize. "She sang that she was 'simply on fire with desire' and that was really her big break."

Soon after, Burnett landed a role in Broadway's "Once Upon a Mattress," and began appearing on morning TV's "The Garry Moore Show."

She never thought she could host her own show. "I was more of a second banana," she said. But she loved playing a variety of characters.

CBS signed her to a 10-year contract doing guest shots on sitcoms and performing in one TV special a year, but the deal also allowed her the option of creating her own variety show and guaranteed her airtime. But five years in, CBS executives had forgotten about the idea.

She recalled one executive telling her: "You know, variety is a man's game."

"At that time, I understood what he was saying, and I was never one to get angry," Burnett said. "I said 'well this is what I know, and this is what I want to do.'"

The show ran from 1967 to 1978 and included guest stars such as Lucille Ball, Jimmy Stewart, Ronald Reagan and Betty White.

Burnett said it's a thrill to receive the award named for humorist and satirist Mark Twain and that she's in good company with past honorees, who include Fey, Bill Cosby, Steve Martin, Lily Tomlin and Ellen DeGeneres.

Coming on the heels of the government shutdown, Burnett joked that she hopes the Kennedy Center's doors will open "so we can get in."

McGarr said it's nice to bring an "intentionally funny moment" to Washington after weeks of political drama.

"You know, serious times call for seriously funny people," McGarr said.

Burnett made a special request that rising comedienne Rosemary Watson, who does impressions of Hillary Clinton and others, be part of the show. Burnett found Watson on YouTube after receiving a fan letter and thought she was funny.

"The thing is, you pay it forward," Burnett said, "because when I got started, somebody gave me a break when I was 21 years old, and I wanted to go to New York."

Before the show, Watson said that watching Burnett shaped her life as a child. She said Burnett was not a joke teller but created funny characters.

"I do what I do because of her," Watson said. "For me, she was it. She was the female comedian I wanted to be most like."

Vicki Lawrence, a co-star with Burnett on "The Carol Burnett Show," said she was planning to be a dental hygienist before she knew Burnett.

"I was going to be cleaning teeth somewhere, and I guess she changed that," Lawrence said.

Tina Fey also said Burnett was an inspiration because her show was the first such variety show led by a woman that Fey had ever seen. Comedy pal Amy Poehler said more than anything, Burnett is just "crazy funny."

Martin Short said anyone who has done sketch or character comedy has borrowed or stolen from Burnett.

"I think she is just a clown in the greatest sense," he said. "She is a satirist in the greatest sense."

** NOVATEL WIRELESS INC (NasdaqGS: NVTL - news) , $2.96, up 4.6 pct

Patent licensing company Wi-Lan said it entered a
patent license agreement with Hewlett-Packard and settled all
pending patent litigation. Under the agreement, Hewlett-Packard
was granted a license related to some WiLAN patents and HP
wireless products. Financial terms were not disclosed.

Wi-LAN also said it settled all pending litigation with
Novatel Wireless, granting the company license to certain WiLAN
products.

** YAHOO INC (NasdaqGS: YHOO - news) , $33.71, down 1 pct (9:55 a.m. ET)

The company's advertising revenue isn't expected to show
much improvement during the third quarter, but Wall Street will
be looking for signs that the Web portal is making gains growing
its audience and enticing people to spend more time on its
websites. Investors will also home in on the included results of
Chinese e-commerce giant Alibaba, in which Yahoo owns a 24
percent stake, and which is planning for a massive public
offering.

** INTEL CORP, $23.52, up 0.3 pct (9:55 a.m. ET)

When the company reports results, investors will want
evidence that recently appointed Chief Executive Brian
Krzanich's new focus on mobile is leading to concrete results.
The company has promised its new Bay Trail mobile chip would
find its way into a slew of new tablets but it is unclear how
much progress Intel has actually made in mobile and whether the
new chips could also cannibalize sales of more profitable PC
processors.
